Proteinase K is a high-activity serine protease (subtilisin family) purified from Tritirachium album Limber. It efficiently degrades proteins and is commonly used for protein removal in biological samples. Proteinase K exhibits broad substrate specificity, with particular preference for cleaving ester and peptide bonds adjacent to the C-terminus of hydrophobic amino acids, sulfur-containing amino acids, and aromatic amino acids. The product exhibits stable activity in Ca2+ salt solutions, and its activity increases in the presence of protein denaturants such as SDS or urea.
Used in DNA/RNA extraction to degrade proteins and inactivate DNases/RNases.
